| dc.description.abstract | Bu tez çalışmasında biyopolimer nanolif üretim sistemlerinin ve bu malzemelerin mekanik ve optik karakterizasyonuna yönelik cihazların tasarımı, imalatı ve optimizasyonunu başarıyla gerçekleştirmiştir. Çalışma, özellikle biyomedikal uygulamalar için nanolif üretim yöntemleri geliştirmeye ve biyomalzemelerin hassas nicel analizine odaklanmıştır. Çalışma kapsamında, merkezkaç kuvvetine dayalı Santrifüj Eğirme Sistemi tasarlanmış ve üretilmiştir. %20 'lik Polivinil Alkol (PVA) konsantrasyonu ve 12.000 RPM dönüş hızı altında, 362 ± 38 nm ortalama çapı olan düzgün morfolojili nanolifler üretilmiştir. Sistemin jelatin gibi farklı biyopolimerlerle uyumluluğu da kanıtlanmıştır. Malzeme karakterizasyonu alanında, ince biyolojik filmlerin mekanik özelliklerini belirlemek için 0,01 N kuvvet ve 0,48 µm yer değiştirim çözünürlüğüne sahip bir Mikro Çekme Test Cihazı ve gözenekli iskelelerin basma kuvveti altındaki davranışını analiz etmek üzere bir Mikro Basma Test Düzeneği geliştirilmiştir. Bu sistemler, kollajen iskelelerin elastik, plato ve yoğunlaşma bölgelerini hassasiyetle ölçerek mekanik modifikasyonların etkilerini doğrulamıştır. Optik analizler için ise, Mueller Matris Polimetri sistemine entegre edilebilen, 4°C ile 55°C ile arasında kontrol sağlayan bir Sıcaklık Kontrol Sistemi tasarlanmış ve biyomalzemelerin sıcaklığa bağlı optik değişimlerini inceleme olanağı sunulmuştur. Ayrıca, Santrifüj Eğirme yöntemi kullanılarak Karboksimetil Selüloz (CMC) takviyeli PVA kompozit nanolif membranlar üretilmiştir. Karakterizasyon sonuçları, CMC nanokristal kümelerinin varlığının, kompozit nanoliflerin morfolojik, mekanik ve optik özellikleri üzerinde kritik bir role sahip olduğunu ortaya koymuştur. Sonuç olarak, bu çalışma, yüksek voltaj gerektirmeyen, ölçeklenebilir nanolif üretim platformlarının geliştirilmesinde önemli bir ilerleme kaydetmiş ve biyomalzemelerin mekanik ve optik performansını hassasiyetle karakterize etmeye yarayan test cihazlarını başarıyla imal etmiştir. Elde edilen bulgular, bu tekniklerin doku mühendisliği, ilaç salım sistemleri ve ileri filtrasyon uygulamaları için nanolif üretimine yönelik güçlü alternatifler olduğunu kanıtlamaktadır. | |
| dc.description.abstract | In this thesis, the design, manufacturing, and optimization of biopolymer nanofiber production systems and devices for the mechanical and optical characterization of these materials were successfully achieved. The study primarily focused on developing nanofiber production methods for biomedical applications and the precise quantitative analysis of biomaterials. Within the scope of the study, a Centrifugal Spinning System based on centrifugal force was designed and produced. As a result of optimization, uniform morphology nanofibers with an average diameter of 362 ± 38 nm were produced under a 20% Polyvinyl Alcohol (PVA) concentration and a rotation speed of 12,000 RPM. The system's compatibility with different biopolymers such as gelatin was also proven. In the field of material characterization, a Micro Tensile Testing Device with 0.01 N force and 0,48 µm displacement resolution was developed to determine the mechanical properties of thin biological films, and a Micro Compression Testing Setup was developed to analyze the behavior of porous scaffolds under compression force. These systems precisely measured the elastic, plateau, and densification regions of collagen scaffolds, validating the effects of mechanical modifications. For optical analyses, a Temperature Control System that can be integrated into the Mueller Matrix Polarimetry system, providing control between 4°C and 55°C, was designed, offering the opportunity to examine temperaturedependent optical changes of biomaterials. Additionally, PVA composite nanofiber membranes reinforced with Carboxymethyl Cellulose (CMC) were produced using the Centrifugal Spinning method. Characterization results revealed that the presence of CMC nanocrystal clusters played a critical role in the morphological, mechanical, and optical properties of the composite nanofibers. In conclusion, this study has made significant progress in the development of scalable nanofiber production platforms that do not require high voltage and has successfully manufactured testing devices for precisely characterizing the mechanical and optical performance of biomaterials. The obtained findings demonstrate that these techniques are strong alternatives for nanofiber production in tissue engineering, drug delivery systems, and advanced filtration applications. | |
