Kanser Hücrelerinde Serin-Glisin Amino Asit Kısıtlamasının Moleküler İncelenmesi ve LC-MS/MS Yöntemi ile Metabolit Analizi

Kanser hücrelerinde metabolik yolaklar enerji üretimini artıracak şekilde yeniden düzenlenir. Kanser hücrelerinin metabolizmalarında rol alan biyokimyasal yolakların aydınlanmasına karşın farklı fizyolojik durumlarda bu yolaklarda meydana gelen değişimlerin nasıl düzenlendiği halen bir araştırma konusudur. Ön sonuçlarımızda serin ve serin/glisin içermeyen besiyerilerinde çoğalamayan hücrelerde ?-glutamil-hidrolaz (GGH) ekspresyonunun olmayışı bize GGH ekspresyonunun serin metabolizmasında da rolü olabileceğini düşündürmüştür. Bu çalışmada GGH ekspresyonun serin besin kısıtlamasındaki rolünü ve terapötik anti-folatlara verilen yanıtı in vitro koşullarda araştırılması amaçlandı. GGH knock-out (KO) ve overeksprese (OE) izojenik hücre hatları elde ederek, serin, glisin ve serin/glisin içermeyen ortamda hücre çoğalmalarındaki farklılıklar hücre canlılık deneyi ile araştırıldı. GGH için izojenik hücre hatlarının 4 farklı besiyerinde metotreaksat duyarlılıkları incelendi. PHGDH inhibitörü CBR-5884 ile serin endojen sentez inhibisyon deneyleri ile ekzojen ve endojen serin olmayışının hücre canlılığına etkisi değerlendirildi. Serin-glisin metabolizması ile ilgili bazı genlerin ekspresyonlarına bakıldı ve LC-MS/MS ile GGH KO hücre hatlarında 4 farklı besiyerinde metabolit analizi yapıldı. Hücre canlılık seviyeleri serin bağımlı kontrol hücrelerine göre GGH KO hücre hatlarında serin içermeyen besiyerinde istatiksel olarak anlamlı bir şekilde fazla idi. Serin bağımlı K562 GGH KO hücre hatlarında hücre canlılığını destekleyecek şekilde kontrol grubuna göre PHGDH gen ekspresyonu anlamlı olarak yüksek bulundu. CBR-5884 inhibitörü GGH KO olan hücrelerde hücre canlılığı anlamlı olarak daha da azalttı. Metabolit analizlerinde GGH KO hücrelerde kontrole göre metabolitlerin anlamlı olarak daha az olduğu bulundu. Bulgularımız sonucunda; serin bağımlı hücrelerde GGH'ın serin-glisin metabolizmasında rolü olduğunu gösterdik. Bu hücrelerde GGH geninin silinmesi serin de novo sentezini arttırmıştır. Folatın hücre dışına gönderilememesi ve hücre proliferasyonunun artması onkojenik bir etki yaratmıştır. Anahtar Sözcükler: Kanser, GGH, Serin-Glisin, Metotreaksat, Metabolit
In cancer cells, metabolic pathways are reorganised to increase energy (ATP) production. Although the biochemical pathways involved in the metabolism of cancer cells have been elucidated, how these pathways are regulated in different physiological conditions is still a subject of research. In our preliminary results, the absence of GGH expression in cells that did not proliferate in serine and serine/glycine-free media suggested that GGH expression may also play a role in serine metabolism. In this study, we aimed to investigate the role of GGH expression in serine nutrient restriction and the response to therapeutic anti-folates in vitro. GGH knock-out (KO) and overexpressed (OE) isogenic cell lines were obtained and the differences in cell proliferation in serine, glycine and serine/glycine-free medium were investigated by cell viability assay. Methotrexate sensitivity of isogenic cell lines for GGH was analysed in 4 different media. The effect of the absence of exogenous and endogenous serine on cell viability was evaluated by serine endogenous synthesis inhibition experiments with the PHGDH inhibitor CBR-5884. The expression of some genes related to serine-glycine metabolism was examined and metabolite analysis was performed by LC-MS/MS in GGH KO cell lines in 4 different media. Cell viability levels were statistically significantly higher in serine-free medium in GGH KO cell lines compared to serine-dependent control cells. PHGDH gene expression was significantly higher in serine-dependent K562 GGH KO cell lines compared to the control group, supporting cell viability. CBR-5884 inhibitor significantly further reduced cell viability in GGH KO cells. Metabolite analyses showed that metabolites were significantly lower in GGH KO cells compared to control. As a result of our findings; we showed that GGH has a role in serine-glycine metabolism in serine-dependent cells. Deletion of the GGH gene in these cells increased serine de novo synthesis. The inability to send folate out of the cell and increased cell proliferation created an oncogenic effect.
