Alzheimer hastalığı in vitro modelinde epps ve nac-nanopartikülü kombine etkisinin araştırılması

| dc.description.abstract | Alzheimer hastalığı (AH), ilerleyici bilişsel kayıplara neden olan ve kesin tedavisi bulunmayan nörodejeneratif bir hastalıktır. Bu çalışmada, AH patofizyolojisinde rol oynayan amiloid beta oligomerleri ile (Aβ42o) toksisitesiyle oluşturulmuş in vitro modelde, 4-(2-hidroksietil)-1-piperazinpropan sülfonik asit (EPPS) ve N-asetilsistein (NAC) yüklü polimer nanopartiküllerin (NAC-NP) monoterapötik ve co-terapötik etkileri değerlendirilmiştir. Hücre canlılığı, mitokondriyal membran potansiyeli, oksidatif stres ve sitokin seviyeleri gibi parametreler üzerinden terapötik etkinlik belirlenmiştir. JC-10 analizi ile mitokondriyal disfonksiyon, ELISA ile IL-1β düzeyi incelenmiştir. Ayrıca, SOD aktivite ve antioksidan ölçümleriyle oksidan savunma mekanizması değerlendirilmiştir. SynergyFinder analizinde Bliss modeline göre antogonist olmayan, nötre yakın edilmiştir. Yapılan analizlerden elde edilen bulgular, EPPS ve NAC kombinasyonunun sinerjik bir etki gösterdiğini ve Aβ42o kaynaklı toksisiteyi azalttığını göstermiştir. B6 ve QSH peptidleri ile fonksiyonellendirilen NAC-NP'nin kan-beyin bariyerini geçtiği ve amiloidleri hedeflediği in vivo koşullarda gözlenmiştir. NAC-NP'nin antiinflamatuar ve antioksidan etkileri gözlemlenirken, nanopartikülün sağladığı kontrollü salım ve EPPS'nin antiamiloid etkisi ile kombine uygulama mitokondriyal bütünlükte iyileşme sağlamıştır. Bu sonuçlar, kombine tedavinin AH'ye yönelik potansiyel bir yaklaşım olabileceğini ve gelecekte yapılacak in vivo çalışmalarla desteklenmesi gerektiğini göstermektedir. | |
| dc.description.abstract | Alzheimer's disease (AD) is a neurodegenerative disorder that causes progressive cognitive decline and currently has no definitive treatment. In this study, the mono-therapeutic and co-therapeutic effects of 4-(2-hydroxyethyl)-1-piperazinepropanesulfonic acid (EPPS) and N-acetylcysteine (NAC)-loaded polymeric nanoparticles (NAC-NP) were evaluated in an in vitro model established by the toxicity of amyloid beta oligomers (Aβ42o), which play a role in AD pathophysiology. Therapeutic efficacy was determined based on parameters such as cell viability, mitochondrial membrane potential, oxidative stress, and cytokine levels. Mitochondrial dysfunction was assessed using JC-10 analysis, and IL-1β levels were measured by ELISA. In addition, antioxidant defense mechanisms were evaluated through SOD activity and antioxidant measurements. SynergyFinder analysis using the Bliss model indicated a non-antagonistic, near-neutral interaction. The results obtained from these analyses showed that the combination of EPPS and NAC exhibited a synergistic effect and reduced Aβ42o-induced toxicity. Under in vivo conditions, it was observed that NAC-NP functionalized with B6 and QSH peptides crossed the blood–brain barrier and targeted amyloid structures. While the anti-inflammatory and antioxidant effects of NAC-NP were observed, the combined application of the nanoparticle's controlled release and the anti-amyloid effect of EPPS's resulted in improved mitochondrial integrity. These findings suggest that combination therapy may be a potential approach for AD and should be supported in future in vivo studies. | |
