dc.description.abstractAnkilozan spondilit (AS), aksiyal spondiloartropatiler grubundan; kronik, otoimmün bir inflamatuar artrittir. AS, birincil olarak omurga ve kuyruk sokumu eklemlerini tutarak sırt ağrısı, hareket kısıtlığı ve omurga kaynaşmasına neden olmakla birlikte, vücuttaki diğer eklemleri ve göz, bağırsak ve deri gibi organları da etkilemektedir. Dünyada AS görülme sıklığı yaklaşık %1'dir. HLA-B27 geni, AS oluşumunun %80-90'ından sorumlu olmakla beraber, HLA-B27 genine sahip insanların sadece %5'i AS geliştirmektedir. Ayrıca, HLA-B27 geninin AS oluşumu nasıl başlattığı da bilinmemektedir. Bu çalışma, AS hastalığının moleküler mekanizmasının daha iyi anlaşılabilmesi ile teşhis ve tedaviye yönelik yeni biyobelirteçlerin bulunabilmesi amacıyla gerçekleştirilmiştir. Bu bağlamda, çeşitli seviyelerdeki -omik verilerin entegre analizi gerçekleştirilmiştir. Transkriptomik çalışmalar kapsamında üç ayrı veri seti üzerinde diferansiyel gen ekspresyonu (DEG) ve ağırlıklandırılmış gen birlikte anlatım ağ analizi (WGCNA) gerçekleştirilmiştir. Epigenomik çalışmalar için diferansiyel metile gen (DMG) tespiti yapılmıştır. Regülomik çalışmalar kapsamında difreansiyel metile gen promotörlerine bağlanan transkripsiyon faktörleri (TF) ile bunların hedef genleri tespit edilmiştir. En son aşamada ise promotörü diferansiyel metile olmuş genler, bu genleri kontrol eden TF'ler ve bu TF'lerin hedefleri kullanılarak bağlama özel bir protein-protein etkileşim ağı (PPI) kurulmuş ve transkriptomik veriler bu ağ üzerine haritalanarak epigenetik ve diğer düzenleyici faktörlerin etkisi altında ekspresyon seviyesi değişen genler tespit edilmiştir. Buna göre tespit edilen genler, AS hastalığının moleküler mekanizmasına daha fazla ışık tutabilecek olup, teşhis ve tedavide yeni biyobelirteçler olarak önerilebilir.
dc.description.abstractAnkylosing spondylitis (AS) is a member of axial spondyloarthropathies and it is a chronic, autoimmune inflammatory arthritis. AS primarily affects the spine and sacroiliac joint causing back pain, limitation of movement and vertebral joint fusion. It also affects other joints in the body and organs such as eyes, intestine and skin. The prevalence of AS in the world is approximately 1%. Although the HLA-B27 gene is responsible for 80-90% of AS, only 5% of people with the HLA-B27 gene develop AS. Furthermore, how the HLA-B27 gene initiates AS is still unknown. This study was carried out in order to better understand the molecular mechanism of AS and to find new biomarkers for diagnosis and treatment. In this context, integrated analysis of -omic data at various levels has been conducted. Within the scope of transcriptomic studies, differential gene expression (DEG) and weighted gene co- expression network analysis (WGCNA) were performed on three separate data sets. Differential methylated gene (DMG) detection was performed for epigenomic studies. For regulomic studies, transcription factors (TF) binding to differential methylated gene promoters and their target genes were determined. Lastly, a context- specific protein-protein interaction network (PPI) was established by using differentially methylated genes, the TFs controlling these genes, and the targets of TFs, and transcriptomic data were mapped onto this network to find the genes that are differentially expressed due to control by epigenetic and other regulatory mechanisms. Accordingly, the detected genes may shed more light on the molecular mechanism of AS and may be recommended as new biomarkers in diagnosis and treatment.
